摘要:
为了筛选桐花树叶75%乙醇提取物乙酸乙酯部位的抗氧化与抗过敏活性组分,通过硅胶柱层析、薄层层析分离出该部位的组分,采用DPPH自由基清除体系、还原力测定体系检测各组分的抗氧化活性,体外透明质酸酶抑制实验检测各组分的抗过敏活性.结果表明:相比于其他组分,组分Fr.3具有较强的DPPH自由基清除能力与还原力,DPPH自由基的半抑制浓度IC50为0.09 mg/mL,远小于人工合成抗氧化剂2,6-二叔丁基对甲酚(BHT)的IC50(0.30 mg/mL); 还原力的半
Abstract:
To screen the antioxidant and antiallergic active fractions of ethyl acetate extract from 75% ethanol extract of Aegiceras corniculatum leaves,the fractions were separated from ethyl acetate extract by silica gel column chromatography and thin layer chromatography(TLC),the antioxidant activities of each fraction were determined by the assays system of 1,1-diphenyl-2-picrylhydrazyl(DPPH·)radical and reducing power measurement system,and the antiallergic activities of each fraction were determined by the ecto-suppress hyaluronidase experimentation.The results show that the component of Fr.3 has stronger DPPH radical scavenging ability and reducing power than other components,its IC50 value of DPPH radical scavenging ability is 0.09 mg/mL,which is far less than butylated hydroxytoluene value of 0.30 mg/mL,its EC50 value of reducing power(0.25 mg/mL)is less than that of Vit.C(0.91 mg/mL)obviously.It also is found that the components of Fr.2,Fr.5,Fr.9 have strong antiallergic activities,with IC50 value of 0.53 mg/mL,0.16 mg/mL,0.25 mg/mL,respectively,which is better than or equivalent to dipotassium glycyrrhizinate value of 0.54 mg/mL.It is thus clear that the strong antioxidant activity component from ethyl acetate extract of A.corniculatum leaves is the component of Fr.3,and the antiallergic activity components are Fr.2,Fr.5,Fr.9.
