摘要:
分别以热水法提取鼠尾藻多糖提取物,超声波辅助-乙醇提取法提取鼠尾藻乙醇提取物.测定鼠尾藻多糖提取物的理化性质、DPPH·自由基清除能力、酪氨酸酶激活活性; 金属离子与食品添加剂对鼠尾藻乙醇提取物DPPH·自由基清除活性的影响.结果表明:鼠尾藻多糖提取物多糖含量45.72%,酸解后含有还原性糖; 多糖提取物浓度为3.6 mg/mL时,DPPH·自由基清除率可达80.95%,对DPPH·自由基清除的EC50为1.41 mg/mL; 该多糖提取物对酪氨酸酶具有激活活性.金属离子对鼠尾藻乙醇提取物DPPH·自由基清除能力均具有增效作用,其中,Fe3+、Zn2+相对较大,增效分别达27.21%和38.08%; 食品添加剂中维生素C、香兰素、蔗糖和柠檬酸对鼠尾藻乙醇提取物DPPH·自由基清除能力表现为增效作用,而苯甲酸钠表现为抑制作用.可见,鼠尾藻多糖与乙醇提取物可作为抗氧化剂的来源,多糖提取物有作为酪氨酸酶激动剂的潜在价值.
Abstract:
The crude polysaccharides and ethanol extracts of Sargassum thunbergii were extracted by hot water extraction and ultrasound-ethanol extraction respectively.The physical and chemical properties,free radical scavenging ability and tyrosinase activation activity of polysaccharide extracted from Sargassum thunbergii chinensis were determined by DPPH· method.And the effect of metal ions and food additives on DPPH· free radical scavenging activity of the ethanol extract were determined.The results showed that the polysaccharides extracted from Sargassum thunbergii had reductive polysaccharides after acidolysis reductive polysaccharides.When the concentration of polysaccharide extract was 3.6 mg/mL,the scavenging rate of DPPH· free radical reached 80.95%,and the EC50 of DPPH· free radical scavenging was 1.41 mg/mL.The polysaccharide extract had the activity of tyrosinase activation metal ions have synergistic effect on DPPH· free radical scavenging ability of ethanol extract of Sargassum thunbergii,among which Fe3+and Zn2+are relatively large,with the synergistic effect reaching 27.21% and 38.08% respectively; vitamin C,vanillin,sucrose and citric acid in food additives had synergistic effect on DPPH· free radical scavenging ability of ethanol extract of Sargassum thunbergii,while sodium benzoate showed inhibitory effect.It can be seen that the polysaccharide and ethanol extract of Sargassum thunbergiican be used as the source of antioxidants,and the polysaccharide extract has potential value as a tyrosinase activator.
